This is due to a change (mutation) in a tumor ...WhatisLFS?|Li-FraumeniSyndromeAssociationHomeWhatisLFS?Li-Fraumenisyndrome(LFS)isaninheritedfamilialpredispositiontoawiderangeofcertain,oftenrare,cancers.Thisisduetoachange(mutation)inatumorsuppressorgeneknownasTP53.Theresultingp53proteinproducedbythegeneisdamaged(orotherwiserenderedmalfunctioning),andisunabletohelppreventmalignanttumorsfromdeveloping.Childrenandyoungadultsaresusceptibletodevelopingseveralmultiplecancers,mostnotablysoft-tissueandbonesarcomas,breastcancer,braintumors,adrenocorticalcarcinomaandacuteleukemia.OthercancersseeninLFSpatientsincludegastrointestinalcancersandcancersofthelung,kidney,thyroid,andskin,aswellasingonadalorgans(ovarian,testicular,andprostate.)ItisimportanttonotethatnoteveryonewithaTP53genemutationwillnecessarilydevelopcancer,buttherisksaresubstantiallyhigherthaninthegeneralpopulation.AdiagnosisofLFSiscriticallyimportantsothataffectedfamiliescanseekappropriategeneticcounselingaswellassurveillanceforearlydetectionofcancer. IntroductionLFSwasfirstrecognizedinthe1969byDrs.FrederickLiandJosephFraumeni,Jr.,whilestudyingpediatricandfamilialcancersattheNationalCancerInstitute.Theydescribedfourfamilieswithmultipleearly-onsetcancersinchildrenandyoungadults.Thesyndromewasfirstreportedinapublicationas“Li-Fraumenisyndrome”in1982byresearchersintheUnitedKingdomwhodescribedtwofamilieswithmultipleformsofcancerinyoungpeople.In1990,inheritedvariantsofTP53werediscoveredastheprimarycauseofLFS.Thisfindingprovidedaspecialopportunityforgenetictestingandclinicalinterventionsthatenablecancerprevention,earlycancerdetection,andcancertreatmentofpeoplewithLFS.ThefindingalsofueledfurthermolecularresearchintoTP53whichiscommonlyfoundinthetumortissueofcancerpatients.
